Methodology
Methodology: Indirect Immunofluorescence HEp-2000 Substrate.
If initial test result is 1:160 or greater, a reflex order for testing to Include titer and pattern will be performed. .

Specimen Requirements
Specimen Type: Serum
Container/Tube: Gold top serum gel-Plain,
red-top tube is also acceptable.
Specimen Volume: 5 mL
Specimen Minimum Volume: 1 mL
Collection Instructions:
1. Fasting (preferred).
2. Causes for rejection include hemolyzed or extremely
lipemic specimen.
Specimen Transport Temperature
Refrigerate: 7 days
Frozen: >7 days
Reference Values
≤1:40 is considered normal
>1:160 is abnormal
Values between 1:80 and 1:160 are borderline abnormal.
Interpretation: If fluorescent ANA test is positive, follow up antibody testing can quantitate and specify the type of
antibody as an aid to diagnosis and management. If fluorescent ANA test is negative and the clinical picture suggests Sjögren’s syndrome, progressive systemic sclerosis (PSS), or polymyositis, testing for “marker” antibodies to SS-A, SS-B, Scl l, and PM l, respectively may be indicated.
