Specimen Requirements
Submit only 1 of the following specimens:
Preferred
Specimen Type: Serum
Container/Tube: Gold top serum gel-Plain
red-top tube is also acceptable.
Specimen Volume: 5 mL
Specimen Minimum Volume: 1 mL
Collection Instructions:
1. Fasting (8 hour). Indicate patient’s fasting
status on request form.
2. Fill tube completely excluding any air bubbles.
3. Invert gently several times to mix blood.
4. Cause for rejection includes hemolysis.

Container/Tube: Green top (heparin)
Specimen Volume: 5 mL
Specimen Minimum Volume: 1 mL

Note: there are several specimen handling factors involved inaccurate ionized calcium measurement.
1) High concentrations of heparin can interfere: for this reason Heparinized tubes should be 3/4 full.
2) ICAL is sensitive to pH changes.
- Exposure to air will cause the co2 in the sample to evaporate which will raise the ph. For this reason, tubes must remain capped until tested.
- Prolonged exposure to metabolically active cells will lower the pH, for this reason heparinized
- whole blood should be kept cool, and serum gel tubes should be spun.
